Tsukushi Makino is the main female character in Boys Over Flowers. Tsukushi is from a poor family, yet attends Eitoku Academy, a school for the super-rich. Her parents force her to attend Eitoku Academy so that she may find a rich husband.

One day in her school, she defends her friend who bumped into Tsukasa, the leader of F4 (The Flowery Four), at the staircase by accident. She receives a red slip from the F4 and all of the students in Eitoku Academy start to bully Tsukushi.

At first she has feelings for Rui Hanazawa. Later she slowly falls in love with Tsukasa Domyoji.
