Lust was created by Scar's brother when he attempted to use alchemy to bring his lover back from the dead, and it is speculated that during the transmutation he lost part or all of his male reproductive organs as part of the equivalent exchange. She was not the first Homunculus to bear the name of Lust, and is the third youngest Homunculi, with only Wrath and Sloth being younger. There are a number of occasions she shows snippets of kindness and sympathy, particularly towards Scar and the Elrics near series' end. No longer trusting Dante, she betrayed Wrath and forms an allegiance with Edward, under the condition that he and Alphonse use the Philosopher's Stone to help achieve her goal of becoming human. These dreams would not come to pass, as Wrath immobilizes and kills her with the help of a locket containing strands of hair from the woman she was originally created from. At the end Lust is left to wonder - was the true reason she so desired to become human was so she could die?

He was created by Hohenheim when he attempted to use alchemy to revive his and Dante's son, who had died from mercury poisoning when he was eighteen years old, suggesting that (as mercury is a key component of alchemy) Hohenheim's research may have been the cause of his death. Envy held a great hatred toward Hohenheim for never being acknowledged by the man as his son and for abandoning him and Dante, with whom he had a rocky relationship. But by the time he learned that Hohenheim remarried and fathered two sons, Envy was even more furious and took it out on his younger half-brothers, Ed and Al, as it was the next best thing to killing their father. He succeeds in killing Ed, though Al managed to revive him with the help of the Philosopher's Stone. Upon learning Hohenheim was alive and beyond the Gate, Envy - still obsessed with the thought of killing him - is able to successfully open and make his way through the doors, changing into Edward, his original self, then a massive serpent

In the anime as the youngest Homunculus, Sloth had become somewhat of a key member of the group. She assumed the identity of Captain Juliet Douglas, a soldier who was said to have sparked the Ishbal War by killing an unarmed Ishbal child (yet mysteriously died two years before the war had started) and died two years ago during the time the Elrics were looking for the Philosopher's Stone. Through manipulation and plotting with Pride, she became his secretary - serving as his emissary/executive assistant. Sloth is hydrokinetic and is able to convert parts or her whole body to liquid; allowing her to travel rapidly through water, and use water based attacks such as drowning her victims with her limbs and making a spinning waterspout to attack. She was created by the Elric brothers when they tried to resurrect their mother, Trisha Elric, though they did not learn of this until much later. For this reason, she looks nearly exactly like her, using it to her advantage on Alphonse until she fell victim to the memories of her "life" as Trisha, seeing that killing the Elric brothers should stop the pain.

Greed was created by Dante. Greed and Envy were bitter rivals, probably due to their connections to Dante, and Envy tormented Greed to the best of his ability. Greed spent the last 140 years imprisoned in the 5th Laboratory by Envy and Dante after Greed betrayed them. The seal that imprisoned Greed was stabilized by the skull of the man Greed himself was modeled after. But when that skull was jarred loose by the explosion caused by Barry's escape from Al and Scar, the seal was broken and Greed was set free. Greed took the opportunity to liberate and recruit the beastmen Chimeras that were imprisoned with him, in order to gain some allies with which to battle in the inevitable confrontation with the other homunculi who he initially assumed were Envy and Gluttony. He promised the half-human creatures sanctuary and revenge on the State Military for what they had done to them. Zolf J. Kimblee and Shou Tucker were in Greed's inner circle as well. He and his followers established their headquarters in the Devil's Nest Bar in the Dublith area, close to where Dante lived.

Wrath was created when Izumi Curtis attempted to bring her child's body back to life with alchemy on Yok Island. The result was a homunculus with its typical inhuman appearance when immature. Izumi sent it back into the Gate where Wrath remained, aging there in the process. When Edward attemped to revive his mother, Wrath took possession of his lost limbs within the gate. The limbs eventually gave Wrath the power to force open the gate and escape back to Yook Island where he lived in the wild for an undetermined length of time.After Sloth's death, Wrath started talking about using the Philosopher's Stone to revive her. Given that Wrath had the ability to perform Alchemy, Dante reacted by summoning the gate. The gate pulled in the human parts of Wrath as it had previously pulled in Edward and his father. For unclear reasons, Wrath could not replace his human limbs with non-human limbs. Winry eventually fitted Wrath with automail similar to Edward's.His whereabouts for the next two years are unknown, though he did resurface to visit his creator's tomb, but appears as a shell of his former self: likely because he has no red stones to consume and accepting who he was after the events of the finale. He guided Alphonse Elric through the underground city to find and open the Gate. Gluttony, ravenous with hunger, attacks, attempting to devour them both. After Gluttony expels some red stone material, Wrath consumes it and regains considerable strength. Wrath is able to hold Gluttony off for a short while, and screams for Al to use both he and Gluttony as Equivalent Exchange to open the portal. Al is wary of sacrificing Wrath, but is assured by the homunculus that that was the very reason he brought Al there in the first place. Upon his redemption we see a human-looking Wrath, standing in front of the doors of the Gate, which open with Izumi there to greet him. Reunited at last, the two disappear into the hereafter together. Wrath's tattoo is on the sole of his right foot.

Physically aging enabled Pride to live among humans and even start a family of his own, with an adopted son named Selim (sometimes translated as Salem), whom he strangled to death when the boy inadvertantly gave Roy Mustang an advantage against him. What Selim thought to be his father's most prized possession was actually the skull of the man Pride was modeled after, who seems similar in appearance to be the first body into which Dante transferred Hohenheim's soul into (Episode 44). However, this also conflicts with Pride's ability to age, causing much controversy . Mustang used the skull to immobilize Pride before repeatedly incinerating him with his alchemic flames, until at length Pride's regenerative ability - powered by the incomplete Philosopher's Stones inside him - was exhausted and he eventually died.
