I own 3 seasons of Rurouni Kenshin but ive only watched 1 1/2 seasons. just after shishio, i have watched about 10-15 episodes.
There is more but i dunno what happens in upcoming eps. sorry

Enishi is Kenshin's brother in law, not his step brother.
The only arc from season three, I'd bother with is Shimabara because of the historical aspect.

There were two novels written. One is called Voyage to the Moon World and I forget the other. They're like the Meiji Arc, you can take them or leave them. They're not part of the canon time line.
